Clinical Description (Database Summary)

(Hérité de SAXAGLIPTINE BIOGARAN 5 mg) In guinea pig models : Saxagliptin (a DPP-4 inhibitor antidiabetic agent) induces a direct, prolongation of the cardiac corrected QT interval and myocardial dysfunction. At clinical and supratherapeutic concentrations it prolongs action potential duration and the QTc interval. Additionally, saxagliptin alters intracellular Ca2+ handling by impairing CaMKII, SERCA2a, and NCX1 functions, leading to diastolic Ca2+ overload and electromechanical systolic/diastolic dysfunction. This off-target cardiotoxicity significantly amplifies risks in patients with pre-existing heart failure or renal impairment due to drug accumulation. However, human trials have not shown an increase in QTc interval when using doses up 400mg daily (80 times the normal dose), according to EMA Caution is advised
