Active Ingredient: PRUCALOPRIDE
Prucalopride (a selective high-affinity 5-HT4 receptor agonist used in chronic constipation) demonstrates no clinically significant effects on cardiac repolarization or QTc intervals. In a thorough QT clinical study of 120 healthy volunteers, therapeutic (2 mg once daily) and supratherapeutic (10 mg once daily) doses showed mean placebo-corrected QTc differences strictly below 5 ms (maximum 3.83 ms), with upper 90% confidence limits below 10 ms. Unlike older non-selective prokinetics such as cisapride, prucalopride lacks hERG channel affinity. A minor transient heart rate increase (up to 5.8 bpm) was documented.
