Clinical Description (Database Summary)

Antifungal triazoles are significantly associated with Torsade de Pointes (TdP) and QT prolongation, with an incidence rate of approximately 2% among reported adverse events. A disproportionality analysis of 18,940 patients revealed a strong association (Reporting Odds Ratio, ROR=7.67). Isavuconazole showed the lowest association (ROR=2.20) but remained statistically significant. Key risk factors include female gender, intravenous (IV) administration (OR=3.08), and specific infection sites such as the central nervous system or heart/bloodstream. The occurrence of these cardiac events doubled the risk of death or life-threatening outcomes (OR=2.03). Concomitant use of QT-prolonging drugs (e.g., Ondansetron, Amiodarone, Levofloxacin) further exacerbates this risk.
