Active Ingredient: BUPRENORPHINE
Transdermal buprenorphine exhibits a dose-dependent effect on cardiac repolarization. In two randomized controlled thorough QT clinical trials evaluating 10, 40, and 80 mcg/h doses, the standard therapeutic dose of 10 mcg/h showed no clinically meaningful QTcI prolongation (upper 90% CI bounds under 10 ms). Supratherapeutic doses of 40 and 80 mcg/h produced QTcI lengthening comparable to 400 mg moxifloxacin (upper bounds reaching 13.3 and 14.1 ms, respectively), which was reversible by naltrexone.
