Active Ingredient: BRENTUXIMAB
Brentuximab vedotin (a CD30-directed antibody–drug conjugate) does not affect cardiac repolarization. In a Phase 1 open-label QTc study of 52 patients with CD30-positive malignancies treated at the standard dose of 1.8 mg/kg, no clinically meaningful effect on ventricular repolarization was found. The upper bound of the 90% confidence interval for the mean change in QTcF remained consistently below 10 ms at all timepoints, including post-infusion (peak parent drug exposure) and Days 2–4 (peak exposure of the free cytotoxic payload, MMAE). In vitro patch-clamp assays confirmed that MMAE does not inhibit hERG $K^+$ channels at clinical levels.
