Clinical Description (Database Summary)

Avacopan is a selective complement C5a receptor antagonist used in ANCA-associated vasculitis that demonstrates no clinically significant effect on cardiac repolarization or QTc intervals. In a phase 1 thorough QTc study evaluating therapeutic (30 mg b.i.d.) and supratherapeutic (100 mg b.i.d.) doses in healthy participants with moxifloxacin positive control, concentration-QTc modeling formally excluded a placebo-corrected QTcF change greater than 10 ms. Mean ddQTcF estimates were 1.5 ms (90% CI -0.17 to 3.09) and 0.8 ms (90% CI -2.41 to 4.05), respectively, with no effect on PR or QRS intervals.
