Clinical Description (Database Summary)

Anagrelide acts as a phosphodiesterase III (PDE III) inhibitor, leading to a dose-dependent increase in heart rate. Its effect on repolarization is nuanced: while some studies show a moderate increase in QTc (up to 15ms) associated with rapid tachycardia, others report that the QTc remains stable while the absolute QT interval shortens due to the increased heart rate. Although the pro-arrhythmic risk is considered low in healthy subjects, caution is strongly advised in patients with pre-existing risk factors (congenital LQTS, electrolyte imbalances, or bradycardia) due to its potent hemodynamic and inotropic effects.
